The study evaluated the prognostic impact of mutations in the SRSF2 gene in patients with acute myeloid leukemia (AML). SRSF2 mutations have been linked to poor outcomes in myelodysplastic neoplasms. This research aimed to assess the impact of these mutations on AML prognosis, noting that SRSF2 mutations are associated with worse overall survival in AML patients. The study analyzed clinical and pathological characteristics of AML patients with SRSF2 mutations and compared them to those with SRSF2 wild-type myeloid neoplasms.<br /><br />Using next-generation sequencing, 134 patients with SRSF2 mutations were identified, including 85 with AML and 49 with non-AML myeloid neoplasms, in addition to 342 SRSF2-wild type AML cases. The results showed that patients with altered SRSF2 exhibited morphologic dysplasia and specific immunophenotypic changes. Additionally, certain gene mutations such as IDH1/2, ASXL1, RUNX1, and STAG2 were more prevalent in SRSF2-mutant AML compared to SRSF2 wild-type cases.<br /><br />Furthermore, the study found that patients with SRSF2-mutant AML had worse overall survival outcomes, but the negative impact on survival could be mitigated by undergoing allogeneic stem cell transplant. The research highlights the importance of considering SRSF2 mutation status in risk stratification and treatment decisions for patients with AML. Future studies with larger sample sizes and multicenter data could provide further insights into the role of SRSF2 mutations in AML prognosis and treatment outcomes.
